SP1 (Phospho-Thr739)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed to detect SP1 protein phosphorylation and expression profile in cells, specifically at the phospho-Thr739 site. It is a convenient, lysate-free, high-throughput assay using an indirect ELISA format in which SP1 (Phospho-Thr739) is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colour-forming reaction on substrate addition. Readout is colourimetric at 450 nm, and the kit is reactive with human, mouse, and rat samples. The stated dynamic range is greater than 5000 cells and the sample type is adherent cells.

To support qualitative interpretation, a monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The kit can be applied to measuring relative amounts of phosphorylated SP1 in cultured cells and to screening the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on SP1 phosphorylation.
