Description
From Assay Genie, this cell-based ELISA kit enables detection of phosphorylated SP1 at threonine 453 in cell samples. SP1 is a transcription factor that plays a critical role in regulating gene expression and cell growth, and phosphorylation at Thr453 has been implicated in modulating its activity in cellular processes including cell cycle progression and apoptosis.
The assay uses an indirect ELISA format: the target is captured by phospho-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction measured at 450 nm. It is described as a convenient, lysate-free, high-throughput and sensitive assay for measuring relative levels of phosphorylated SP1 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on SP1 phosphorylation. The kit shows reactivity with human, mouse and rat samples and has a stated dynamic range of > 5000 Cells. Sample type is adherent cells.
A monoclonal anti-GAPDH antibody is included as an internal positive control to normalize target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density after the colorimetric measurement. SP1 is a C2H2-type zinc finger transcription factor (Gene ID: 6667, UniProt ID: P08047) that is phosphorylated and activated by MAPK, with phosphorylation modulating its activity.
