The Assay Genie SMM ELISA Kit is a competitive inhibition immunoassay built around an antigen-pre-coated microtiter plate and an HRP-labeled conjugate that binds anti-SMM antibody in solution. During the 25°C incubation sequence (45 min, then 15 min after substrate addition), SMM in the sample or standard competes with the plate-bound antigen for antibody binding sites, and TMB substrate produces a color signal whose intensity is inversely proportional to analyte concentration, so higher SMM levels correspond to lower absorbance readings.

Six ready-to-use liquid standards (0, 0.05, 0.15, 0.45, 1.35, and 4.05 ppb) are supplied for plotting the calibration curve. Sensitivity is 0.05 ppb, and matrix-specific limits of detection have been validated as follows: muscle method 1 at 0.05 ppb, muscle method 2 at 0.25 ppb, serum and urine at 0.2 ppb, honey at 0.05 ppb, and milk at 1 ppb. Mean sample recoveries are 95%±25% for muscle and honey, and 85%±25% for urine, milk, and serum. Cross-reactivity is reported at 100% for Sulfamonomethoxine itself, supporting use as a residue-screening tool for veterinary drug monitoring programs.

The microplate kit ships with one HRP conjugate vial (5.5 mL), antibody working solution (5.5 mL), substrate A and B (6 mL each), stop solution (6 mL), 20× concentrated wash buffer (40 mL), and 2× reconstitution buffer (50 mL), plus plate sealers and a foil pouch for returning unused strips. Required lab equipment includes a microplate reader, homogenizer, nitrogen evaporator, water bath, centrifuge, and single- or multi-channel pipettes covering 20-200 uL, 100-1000 uL, and 30-300 uL ranges. Store kit components at 2-8°C, never freeze them, and reseal unused wells in the original foil bag with desiccant; expiry is printed on the outer box (typically 12 months from manufacture). For research use only.
