SMC1 (Phospho-Ser966)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed for the quantitative detection of SMC1 protein phosphorylated at serine 966 in cell samples. The kit uses an indirect ELISA format in which SMC1 (Phospho-Ser966) is captured by a phospho-specific primary antibody, and an HRP-conjugated secondary antibody binds the Fc region of that primary antibody. Addition of a chromogenic substrate produces a colorimetric signal read at 450 nm, allowing qualitative measurement of relative phosphorylation levels in cultured cells with a dynamic range of > 5000 cells.

Validated for Human, Mouse, and Rat samples and intended for use with adherent cells, the kit supports a range of research applications, including screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on SMC1 phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control to normalize target absorbance values, and Crystal Violet whole-cell staining can be used to account for cell density and plating differences. SMC1A has a reported molecular weight of 155 kDa.

SMC1 (Structural Maintenance of Chromosomes protein 1A) is a central component of the cohesin complex, which is required for sister chromatid cohesion after DNA replication and plays roles in DNA repair, spindle pole assembly during mitosis, and the DNA damage checkpoint via ATM and ATR signalling. Phosphorylation at Ser966 is known to regulate cohesin dynamics and chromatin structure, making this kit a relevant tool for studies of cell cycle progression, DNA repair, gene expression, and related disease mechanisms including cancer and Cornelia de Lange syndrome.
