Description

From Assay Genie, this cell-based ELISA kit is intended for the detection and quantification of SMC1 protein levels in cell lysates and tissue homogenates. It uses a colorimetric detection method read at 450 nm and is configured in a 96-well microplate format. The assay is reactive with human and mouse samples, suitable for cell biology, oncology, and genetics research, and has a stated dynamic range of > 5000 Cells. The kit is stored at 4°C for 6 months.

The assay is lysate-free and uses an indirect ELISA format in which SMC1 is captured by SMC1-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control, and a Crystal Violet whole-cell staining step can be used to normalize absorbance values to cell density. The kit can be used to measure relative SMC1 amounts in cultured cells as well as to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on SMC1, providing qualitative determination of SMC1 concentration.

SMC1A (Gene ID: 8243, UniProt: Q14683) is a central component of the cohesin complex, required for the cohesion of sister chromatids after DNA replication and involved in DNA repair through interactions with BRCA1 and phosphorylation by ATM or ATR. Dysregulation of SMC1 has been linked to cancer and developmental disorders, and mutations in the gene have been associated with Cornelia de Lange syndrome. The target protein has a molecular weight of 155 kDa.
