Smad4 Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit enables colorimetric detection of Smad4 protein expression directly in cultured cells using a lysate-free, indirect ELISA workflow. Smad4-specific primary antibodies capture the target within the cells, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm after substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step allows results to be normalized to cell density, compensating for plating differences.

The kit is reactive with human, mouse and rat samples and is intended for qualitative measurement of Smad4 in cultured cells as well as for screening the effects of treatments, siRNA, inhibitors or activators on Smad4 expression. Smad4 functions as a common-mediator (co-Smad) transcription factor of the TGF-β superfamily, binding consensus DNA elements and supporting recruitment of the Smad2/Smad4/Fast-1 complex to target gene promoters. The assay has a dynamic range of more than 5000 cells per well and is designed for adherent cell cultures.
