Smad3 (Phospho-Ser213)Transcription Factor Activity Assay

From Assay Genie, this DNA-binding activity assay quantifies the DNA-binding capacity of Smad3 phosphorylated at Ser213, a key activated form generated downstream of TGF-β receptor signaling. The plate is pre-coated with oligonucleotides carrying the Smad-binding element (SBE) consensus sequence, so only transcriptionally engaged Smad3 is captured from nuclear extracts and detected with a phospho-Ser213-specific antibody.

Typical research uses include mapping TGF-β pathway activation in fibrosis models, epithelial-to-mesenchymal transition (EMT) studies, oncology screening panels, and small-molecule pathway inhibitor profiling in human, mouse and rat samples. Nuclear and cell lysates are loaded directly, with a recommended loading of roughly 5-10 µg of nuclear protein per well.
