Description

This Cell-Based ELISA Kit from Assay Genie is designed to detect the phosphorylation status of Smad2 at Ser467 in cell samples, supporting research into the TGF-beta signalling pathway and related cellular processes including cell growth, differentiation, and apoptosis.

The assay uses a colorimetric detection method read at 450 nm and is reactive with Human, Mouse, and Rat samples. The kit has a dynamic range of > 5000 Cells and detects Smad2 (Phospho-Ser467) qualitatively via an indirect ELISA format, in which target-specific primary antibodies are bound by HRP-conjugated secondary antibodies that catalyse a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density, allowing absorbance values to be adjusted for plating differences.

SMAD2 is a ubiquitously expressed transcription factor that is phosphorylated and activated by TGF-beta receptor-type kinases. Phosphorylated Smad2 dimerises with the collaborating Smad4 and is translocated into the nucleus, where it stimulates the transcription of target genes, participating in morphogenesis, cell-fate determination, proliferation, differentiation, and apoptosis.
