Smad2 Colorimetric Cell-Based ELISA Kit

Assay Genie provides a cell-based ELISA kit for the qualitative detection of Smad2 protein in cultured cells, using a colorimetric readout measured at 450 nm. The kit is supplied in a 96-well microplate format, reacts with human, mouse, and rat samples, and is intended for adherent cell cultures, delivering a lysate-free, high-throughput workflow suitable for measuring relative Smad2 levels and for screening the effects of various treatments, inhibitors, or activators.

Smad2 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary to catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody serves as an internal positive control for normalization, and Crystal Violet whole-cell staining can be applied to determine cell density and adjust for plating differences. The kit has a dynamic range of > 5000 cells and is stored at 4°C for 6 months. It targets SMAD2, a ubiquitously expressed transcription factor in the TGF-β signaling pathway involved in morphogenesis, cell-fate determination, proliferation, differentiation, and apoptosis, with dysregulation linked to cancer, fibrosis, and autoimmune disorders.
