The Smad1 (Phospho-Ser465) Transcription Factor Activity Assay from Assay Genie quantitatively measures the DNA-binding competence of Smad1 specifically when phosphorylated at serine 465, the precise residue targeted by BMP type I receptors (ALK1, ALK2, ALK3, and ALK6). Phosphorylation at this site occurs downstream of bone morphogenetic protein (BMP) ligand engagement and serves as a direct functional readout of active BMP pathway signaling, making the kit especially useful for researchers investigating osteoblast differentiation, chondrogenesis, skeletal development, and BMP-driven stem cell fate decisions.

Smad1 is a receptor-regulated Smad (R-Smad) that, upon BMP-induced phosphorylation, partners with the common-mediator Smad4, accumulates in the nucleus, and binds Smad-binding elements (SBEs) in target gene promoters to regulate transcription of osteogenic and developmental genes. By capturing only the DNA-competent, phosphorylated fraction recovered from nuclear and whole-cell lysates, the assay provides a functional activation endpoint that complements standard phospho-Smad1 western blot workflows. Typical applications include BMP ligand screening, characterization of pathway cross-talk within the TGF-beta superfamily, and interrogation of receptor-specific signaling bias in primary cells and established lines.

For best results, prepare fresh nuclear extracts from treated cultures, store kit reagents at -20 degrees C in a non-frost-free freezer, and protect unused strip wells from moisture. Intended for in vitro research use only.
