Description

Assay Genie's Smad1 Colorimetric Cell-Based ELISA Kit is a high-quality In-Cell ELISA designed for the quantitative measurement of SMAD1 in cell lysates and tissue extracts. The assay uses an indirect ELISA format in which SMAD1 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. Reactivity covers Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells, and the intended sample type is adherent cells.

Provided in a 96-Well Microplate format, the kit is suited for measuring relative SMAD1 amounts in cultured cells and for screening the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on SMAD1. Because the assay is qualitative, two normalization methods are supplied: a monoclonal anti-human GAPDH antibody acts as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density, after which absorbance values can be normalized to cell amounts to adjust for plating differences. Storage is at 4°C with a stability of 6 Months.

SMAD1 is a signal transducer and transcriptional modulator of the TGF-beta / BMP signaling pathway, phosphorylated and activated by BMP type 1 receptor kinases, and participates in morphogenesis, cell-fate determination, proliferation, differentiation and apoptosis. Dysregulation of SMAD1 has been linked to cancer, fibrosis and developmental disorders. Annotated database entries for the target include Gene ID 4086, UniProt ID Q15797, OMIM 601595, Unigene Hs.604588, gene symbol SMAD1, and chromosomal location of the human ortholog 4q31.
