Built as a competitive ELISA for veterinary drug residue screening, this Assay Genie kit delivers quantitative detection of streptomycin in muscle, honey, milk, and milk powder, with matrix-specific limits of detection of 4 ppb for muscle, 2 ppb for honey, and 5 ppb for milk and milk powder. The analytical sensitivity is 0.1 ppb (ng/mL), and the six liquid standards supplied with the kit span 0.1 ppb to 8.1 ppb, generating a six-point calibration curve that produces colorimetric readouts on a standard microplate reader.

Because streptomycin is regulated alongside its close analogue dihydrostreptomycin in most jurisdictions, the antibody cross-reactivity profile is a defining performance feature: streptomycin 100%, dihydrostreptomycin 100%, clarithromycin 6.3%, and gentamicin 2.5%. This dual recognition supports reporting of the combined streptomycin/dihydrostreptomycin residue required by Codex Alimentarius MRLs, including 600 µg/kg in cattle muscle and 200 µg/kg in milk. Sample preparation is matrix-dependent: muscle is defatted with N-hexane and clarified; honey is diluted directly; and milk powder is reconstituted and diluted. After the appropriate dilution in the supplied 5× reconstitution buffer, a 50 μL aliquot is loaded per well, with documented dilution factors of 40 for muscle, 20 for honey, and 50 for milk and milk powder. Validated sample recovery is 85% ± 15% across these matrices.

Each 96-assay microplate kit from Assay Genie includes a pre-coated microtiter plate, HRP conjugate (11 mL), antibody working solution (5.5 mL), substrate reagents A and B (6 mL each), stop solution (6 mL), 20× concentrated wash buffer (40 mL), 5× reconstitution buffer (50 mL), three plate sealers, a foil bag with desiccant, and a printed manual. The reaction proceeds at 25°C in three timed incubations totalling 75 minutes (30 min, 30 min, 15 min). Unused microwells must be returned to the foil bag with desiccant and stored refrigerated; refrigerated shelf life is 6 months from the date printed on the kit label. The kit is intended for research use only and for in vitro food residue analysis, and is not validated for clinical diagnostic use.
