Description

Designed for the quantitative measurement of SKP2/p45 in cell lysates, this cell-based ELISA kit from Assay Genie provides a high-throughput, sensitive assay with reactivity to human samples. The kit uses a colorimetric detection method at 450 nm in a 96-well microplate format and is suited to measuring the relative amounts of SKP2/p45 in cultured adherent cells as well as screening the effects of various treatments, inhibitors, or activators on the target.

Qualitative determination of SKP2/p45 concentration is achieved through an indirect ELISA format in which SKP2/p45 is captured by specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values. Following HRP activity measurement, Crystal Violet whole-cell staining can be used to determine cell density, allowing absorbance values to be normalised to cell amounts and correcting for plating differences.

With a dynamic range of > 5000 Cells, storage at 4°C, and 6 months stability, the kit is suitable for both academic and pharmaceutical research settings. SKP2/p45 is a key regulator of cell cycle progression, involved in the degradation of cell cycle regulators, and its dysregulation has been linked to various cancers, making it a valuable target for cancer biology and drug development research.
