Description

From Assay Genie, this cell-based ELISA kit is designed for the detection and measurement of Signal Regulatory Protein Alpha1 (SIRP alpha1) in cell lysates and tissue homogenates. It is a lysate-free, high-throughput, sensitive assay that detects the SIRP alpha1 protein expression profile in cultured cells, allowing researchers to measure relative protein amounts and screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on SIRP alpha1.

Reactivity is reported for Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. Detection is colorimetric at 450 nm and the format is a 96-well microplate. The assay uses an indirect ELISA format in which SIRP alpha1 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; the conjugated HRP then catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step is provided to determine cell density, allowing absorbance values to be normalised to cell amounts and plating differences to be adjusted.

Storage is at 4°C with a stated stability of 6 months. The kit supports research into immune regulation and cell signalling, where SIRP alpha1 plays a role in processes such as phagocytosis and inflammation, and where dysregulation has been implicated in cancer, autoimmune disorders, and inflammatory conditions. Associated database identifiers for the target are Gene ID 140885, UniProt ID P78324, and OMIM 602461.
