Description

Assay Genie supplies a Cell-Based ELISA kit designed for the accurate detection of phosphorylated SHP-1 protein at Tyr536 in cell lysates and tissue samples. SHP-1 is described as a critical regulator of cellular signaling pathways that influence immune responses, cell growth, and apoptosis. The kit targets SHP-1 (Phospho-Tyr536) and is reactive with Human, Mouse, and Rat samples, with adherent cells indicated as the sample type.

Detection is colorimetric and read at 450 nm, with a stated dynamic range of > 5000 Cells. The assay uses a lysate-free, high-throughput indirect ELISA format in which SHP-1 (Phospho-Tyr536) is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. It can be used to measure relative levels of phosphorylated SHP-1 in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on SHP-1 phosphorylation.

For normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control. Following the colorimetric read, a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalized to cell amounts and plating differences to be adjusted. The associated gene is PTPN6, encoding Tyrosine-protein phosphatase non-receptor type 6 with a reported molecular weight of 64 kDa.
