SHP-1 Colorimetric Cell-Based ELISA Kit

Assay Genie's Cell-Based ELISA Kit is designed for the precise measurement of SHP-1 protein levels in cell samples. The kit uses a colorimetric detection method at 450 nm and is reactive with human, mouse, and rat samples, with a stated dynamic range of > 5000 cells. It is supplied in a microplate format and is intended to be stored at 4 °C, where it remains stable for up to 6 months.

This convenient, lysate-free, high-throughput assay provides qualitative determination of SHP-1 in cultured cells via an indirect ELISA format: SHP-1 is captured by SHP-1-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density, allowing the results to be normalized to cell amounts. The kit can be used for measuring relative SHP-1 levels in cultured cells as well as screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on SHP-1, and is applicable to adherent cell samples.

SHP-1, also known as tyrosine-protein phosphatase non-receptor type 6, is encoded by the PTPN6 gene (UniProt P29350; molecular weight 64 kDa). The protein modulates signaling by tyrosine-phosphorylated cell surface receptors such as KIT and the EGF receptor/EGFR, and plays a key role in hematopoiesis, with dysregulation having been linked to autoimmune diseases, cancer, and inflammatory disorders.
