Description

A cell-based ELISA kit from Assay Genie designed for the colorimetric detection of phosphorylated SHIP1 at Tyr1021 in adherent cells. The product is a convenient, lysate-free, high-throughput and sensitive assay that can be used to measure SHIP1 protein phosphorylation and expression profiles in cultured cells, and to screen the effects of various treatments, inhibitors (e.g. siRNA or chemicals) or activators on SHIP1 phosphorylation. Reactivity is reported for human, mouse and rat samples, with a stated dynamic range of > 5000 cells and detection carried out at 450 nm.

SHIP1 is described as a critical regulator of cellular signalling pathways, playing a key role in modulating immune responses, cell survival and proliferation. Phosphorylation at Tyr1021 is known to impact its enzymatic activity and downstream signalling events, making this phospho-site a valuable marker for studying intracellular signalling cascades, elucidating the protein's role in disease pathology, and evaluating potential therapeutic interventions. The target is encoded by the INPP5D gene (Gene ID: 3635; UniProt ID: Q92835).

Qualitative determination of SHIP1 (Phospho-Tyr1021) is achieved via an indirect ELISA: SHIP1 (Phospho-Tyr1021) is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing a colorimetric readout. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may subsequently be used to determine cell density and adjust for plating differences.
