SHIP1 Colorimetric Cell-Based ELISA Kit

A colorimetric cell-based ELISA kit from Assay Genie provides qualitative detection of SHIP1 protein expression in cultured cells. The lysate-free, indirect ELISA format captures SHIP1 with target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. The kit is reactive with human, mouse, and rat samples, is intended for adherent cell cultures, and is run on a 96-well microplate with absorbance read at 450 nm and a stated dynamic range of more than 5000 cells.

The assay measures relative SHIP1 levels in cultured cells and can be used to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. Because the cell-based ELISA is qualitative, two normalisation strategies are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control for target absorbance values, and Crystal Violet whole-cell staining is used to determine cell density so that absorbance values can be normalised to cell amount. Stated storage stability is 4°C for 6 months.

The target, SHIP1 (gene symbol INPP5D; Gene ID 3635; UniProt ID Q92835), is an SH2-containing inositol phosphatase restricted to hematopoietic cells that hydrolyses Ins(1,3,4,5)P4 and PtdIns(3,4,5)P3 and regulates cell survival, growth, cell cycle arrest, and apoptosis.
