Description

Designed by Assay Genie for sheep sample analysis, this competitive ELISA is coated with antigen and provides a colorimetric detection workflow for quantifying Prostaglandin E2 (PGE2). The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is rated at 18.75 pg/mL with a working range of 31.25-2000 pg/mL, and the kit is stored sealed at 2-8°C (do not cryopreserve).

The protocol follows a standard workflow: equilibrate TMB substrate for 30 minutes at room temperature and pre-wash the plate twice, then add 50 µL of standard or sample with 50 µL of biotin-labeled antibody per well, incubate at 37°C for 45 minutes, wash five times with 1 minute soaks, develop color in the dark at 37°C for 10-20 minutes, stop the reaction (blue to yellow), and read absorbance at 450 nm. Sample preparation includes serum (clot, 1000 × g, 20 minutes), EDTA plasma (1000 × g, 15 minutes at 2-8°C), cell culture supernatant (1000 × g, 20 minutes at 4°C), cell lysates (10,000 rpm, 10 minutes), and tissue homogenates (5000 × g, 5 minutes).

Reported linearity at 1:2, 1:4, and 1:8 dilutions spans 88-102%, 85-101%, and 87-102% for serum; 87-97%, 89-95%, and 82-93% for EDTA plasma; and 85-95%, 82-92%, and 83-98% for heparin plasma (n = 10 each). Mean recovery is 98% for serum (range 92-105%), 97% for EDTA plasma (range 86-103%), and 97% for heparin plasma (range 89-102%). PGE2 is described as a key mediator of inflammation and pain, with dysregulation implicated in arthritis, inflammatory diseases, and cancer, supporting studies of sheep physiology and disease pathology.
