Sheep IL-4 ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the detection of interleukin-4 (IL-4) in sheep samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody sandwich format with colorimetric detection, with absorbance read at 450 nm. Sensitivity is 9.375 pg/mL and the working range is 15.625–1000 pg/mL. The kit is stored at 2–8°C and remains stable for 12 months under those conditions.

The procedure involves equilibrating reagents and TMB substrate to room temperature, then loading standards and samples onto the pre-coated plate and incubating at 37°C for 90 minutes. A biotin-labelled detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is then added and developed in the dark for 10–20 minutes before the reaction is stopped and absorbance is measured at 450 nm.

Reported linearity at 1:2, 1:4 and 1:8 dilutions (n = 5) is 89–104%, 91–104% and 87–104% for serum; 94–99%, 83–92% and 83–98% for EDTA plasma; and 87–95%, 82–97% and 89–95% for heparin plasma. Recovery ranges are 89–104% (average 96%) for serum, 93–105% (average 100%) for EDTA plasma, and 88–104% (average 97%) for heparin plasma.
