Description

A colorimetric Sandwich ELISA, Double Antibody assay from Assay Genie intended for the quantitative measurement of IL-12 (p40) in sheep serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit reports a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL, and the associated Uniprot reference is P68220. The procedure includes a primary incubation at 37°C for 90 minutes, followed by a 60-minute detection antibody step, a 30-minute HRP-Streptavidin binding step, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution. Reagents are stored at 2-8°C for 12 months.

IL-12 p40 is a subunit of interleukin-12, a cytokine that plays a critical role in regulating the immune response and promoting inflammation, with its dysregulation implicated in various inflammatory and autoimmune diseases. Validated sample matrices include serum, EDTA plasma, heparin plasma, and tissue homogenates. Dilution linearity has been reported in serum at 88-102% (1:2), 85-104% (1:4) and 86-102% (1:8), with EDTA plasma at 86-98% (1:2), 85-101% (1:4) and 82-96% (1:8), and heparin plasma at 83-97% (1:2), 85-100% (1:4) and 80-95% (1:8). Reported recovery averages 98% for serum, 98% for EDTA plasma, and 95% for heparin plasma, supporting use of the kit for research applications in immunology and inflammation.
