Description

Assay Genie offers a sandwich double-antibody ELISA kit designed for the quantitative detection of activin A in sheep-derived samples. Built around a colorimetric readout, the kit is intended for research applications including studies of follicle-stimulating hormone production, inflammation, cell differentiation, reproductive disorders, immune dysfunction, and cancer, where activin A serves as a relevant biomarker.

Manufacturer specifications report a sensitivity of 37.5 pg/mL and a working range of 62.5–4000 pg/mL. The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and should be stored at 2-8°C for 12 months. Linearity has been validated at 1:2, 1:4 and 1:8 dilutions for serum (85-98%, 87-93%, 98-105%), EDTA plasma (94-99%, 83-100%, 83-100%) and heparin plasma (81-96%, 81-98%, 83-95%). Spike-and-recovery ranges were 87-102% (average 95%) for serum, 85-101% (average 93%) for EDTA plasma, and 85-102% (average 92%) for heparin plasma. The associated UniProt reference is P43032, and the target is also referred to as ACV-A.

The assay workflow begins with reagent and TMB substrate equilibration, followed by a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step at 37°C, a 30-minute HRP-Streptavidin (SABC) binding step, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution. Sample preparation includes clotting blood for serum (centrifuge at 1000 × g for 20 minutes), collecting plasma with anticoagulant tubes (1000 × g, 15 minutes, 2–8°C), homogenising tissue in PBS with protease inhibitors, clarifying cell culture supernatant at 2500 rpm for 5 minutes, and lysing cells in buffer containing protease inhibitors.
