Description

A cell-based ELISA from Assay Genie designed for the precise measurement of SGK phosphorylation at Ser78, with reactivity against Human, Mouse and Rat samples. The assay uses adherent cells as the sample type and operates on a cell-based, lysate-free, indirect ELISA format in which the target is captured by phospho-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm. The dynamic range is stated as > 5000 Cells.

The kit is intended for measuring the relative amounts of phosphorylated SGK in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on SGK phosphorylation. Because the assay is qualitative, a monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target readings, and a Crystal Violet whole-cell staining method is provided for determining cell density and adjusting for plating differences after the HRP colorimetric read-out.

SGK1 (serum/glucocorticoid-regulated kinase 1) is described as an AGC protein kinase of the SGK family that is rapidly induced by stimuli including serum, glucocorticoids, follicle stimulating hormone, osmotic shock and mineralocorticoids, and that can be activated via PI3K-dependent and -independent pathways. It plays a role in activating certain potassium, sodium and chloride channels and is implicated in processes such as cell survival, neuronal excitability and renal sodium excretion, and is negatively regulated by ubiquitin modification and proteasome degradation.
