SF1 Colorimetric Cell-Based ELISA Kit

Assay Genie offers a Colorimetric Cell-Based ELISA Kit for the detection of Splicing factor 1 (SF1), a nuclear pre-mRNA splicing factor encoded on human chromosome 11q13 (UniProt Q15637; Gene ID 7536). The assay is described as a convenient, high-throughput, sensitive and lysate-free method for measuring relative SF1 protein expression in cultured cells, and for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on SF1 levels.

Reactive with human and mouse samples, the kit is intended for use with adherent cells and detects a dynamic range of > 5000 Cells. Detection is by colourimetric measurement at 450 nm, with components stored at 4 °C for up to 6 months in a microplate format. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising the target absorbance values.

The assay follows an indirect ELISA format in which SF1 is captured by SF1-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colour reaction upon substrate addition. Following this colourimetric read-out, the Crystal Violet whole-cell staining method may be applied to determine cell density, allowing results to be normalised to cell amounts and adjusting for plating differences. The kit is positioned for research applications in epigenetics and nuclear signaling.
