SENP5 Colorimetric Cell-Based ELISA Kit

A cell-based ELISA kit from Assay Genie is designed to measure SENP5 protein expression directly in cultured cells as a convenient, high throughput, and sensitive assay that does not require lysate preparation. SENP5 is a SUMO-specific protease that processes full-length SUMO3 to its mature form and deconjugates SUMO2 and SUMO3 from target proteins, playing a key role in protein sumoylation, desumoylation, and cell division.

The assay uses a 96-well microplate format with a colorimetric detection method measured at 450 nm, and is reactive with human and mouse samples. The kit has a dynamic range of > 5000 Cells, is intended for use with adherent cells, and is suited to cell cycle research. Storage stability is given as 4°C for 6 months.

Detection relies on an indirect ELISA format in which SENP5-specific primary antibodies are captured and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalizing target absorbance, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. Applications include studies of DNA repair, transcriptional regulation, protein stability, cell proliferation, apoptosis, and gene expression.
