SEK1/MKK4 Colorimetric Cell-Based ELISA Kit

A colorimetric cell-based ELISA kit from Assay Genie is designed to detect and measure SEK1/MKK4 protein levels in cultured cells using an indirect ELISA format. The assay is lysate-free, high throughput, and sensitive, making it suitable for studying relative protein expression as well as screening the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on SEK1/MKK4. SEK1/MKK4 is a key signaling molecule in the MAPK signaling pathway, playing a role in cell growth, differentiation, and apoptosis, with dysregulation linked to cancer, inflammatory disorders, and neurodegenerative diseases.

The kit shows reactivity with human, mouse, and rat samples and is intended for adherent cells. Detection is by colorimetric measurement at 450 nm, the dynamic range is greater than 5000 cells, and the format is a 96-well microplate. SEK1/MKK4 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze the colorimetric reaction upon substrate addition. Two normalization methods are included: a monoclonal anti-GAPDH antibody serves as an internal positive control, and Crystal Violet whole-cell staining is used to determine cell density. Storage is at 4°C with a stated stability of 6 months.
