SDCG1 Colorimetric Cell-Based ELISA

From Assay Genie, this colorimetric cell-based ELISA kit is designed for the accurate detection of SDCG1 levels in cell lysates, cell culture supernatants, and tissue homogenates. SDCG1, also known as Syndecan-1, is described as a cell surface proteoglycan with a role in cell adhesion, migration, and signalling, with dysregulation of its expression implicated in cancer, inflammatory disorders, and developmental abnormalities. The kit is reactive with Human, Mouse, and Rat samples and uses a colourimetric detection method at 450 nm.

Qualitative determination of SDCG1 concentration is achieved through an indirect ELISA format, in which SDCG1 is captured by SDCG1-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP enzyme to catalyse a colourimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used after the colourimetric measurement to determine cell density, allowing absorbance values to be normalised to cell amounts. The dynamic range is quoted as > 5000 Cells, the format is a 96-well microplate, and the kit is stable at 4 °C for 6 months.

The assay is lysate-free and high throughput, making it suitable for measuring relative amounts of SDCG1 in cultured cells as well as screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on SDCG1.
