This cell-based ELISA kit from Assay Genie is designed for the detection and quantification of phosphorylated RSK1/2/3/4 at Ser221/227/S218/232 in cell lysates, providing high sensitivity and specificity for research into signalling pathways that regulate cell growth, differentiation, and survival. RSK1/2/3/4 are serine/threonine kinases whose phosphorylation at specific serine residues is essential for activation and downstream signalling, and aberrant phosphorylation has been implicated in cancer, inflammatory disorders, and neurodegenerative diseases.

The kit uses a lysate-free, high-throughput indirect ELISA format. RSK1/2/3/4 (Phospho-Ser221/227/S218/232) is captured by phospho-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction measured at 450 nm. The assay is suited for measuring relative phosphorylation levels in cultured adherent cells and for screening the effects of treatments, inhibitors, or activators. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and Crystal Violet whole-cell staining can be used to assess cell density.
