Description

This cell-based colorimetric ELISA kit from Assay Genie is designed for the qualitative detection of phosphorylated RIPK2 at Ser176 in cultured cells. It is a lysate-free, high-throughput assay based on an indirect ELISA format, in which RIPK2 (Phospho-Ser176) is captured by a target-specific primary antibody and a horseradish peroxidase (HRP)-conjugated secondary antibody catalyses a colourimetric reaction measured at 450 nm. The kit is suitable for measuring phosphorylated RIPK2 levels in adherent cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on RIPK2 phosphorylation.

Reactivity is reported for Human and Mouse samples, with a dynamic range of > 5000 cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalisation of target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. The assay targets receptor-interacting serine/threonine-protein kinase 2, a signalling protein involved in cell death and inflammation pathways whose phosphorylation at Ser176 plays a role in its activation and downstream signalling. The product is supplied as two 96-well microplates and is intended for research applications investigating RIPK2 signalling, inflammation, and related disease mechanisms.
