Description

From Assay Genie comes a Cell-Based ELISA for the qualitative measurement of Receptor-Interacting Serine/Threonine-Protein Kinase 2 (RIPK2) levels in cultured adherent cells. The assay uses a colorimetric detection method read at 450 nm in an indirect ELISA format, where RIPK2 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a color reaction upon substrate addition. Described as a convenient, lysate-free, high-throughput and sensitive kit, it is suited to measuring relative RIPK2 expression and screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on the target.

Reactivity is stated for Human and Mouse samples, and the assay is supplied in a 96-Well Microplate format with a dynamic range of > 5000 Cells. Two normalization strategies are provided: a monoclonal anti-GAPDH antibody acts as an internal positive control, and Crystal Violet whole-cell staining is used to determine cell density so absorbance values can be adjusted for plating differences. Storage is at 4°C for 6 months. The target RIPK2 is a serine/threonine protein kinase of the RIPK family and a component of innate and adaptive immune signaling, capable of activating NF-kappaB and inducing apoptosis.
