Description

A cell-based colorimetric ELISA kit from Assay Genie designed to detect rhodopsin protein expression directly in cultured cells. The kit uses an indirect ELISA format in which rhodopsin-specific primary antibodies are bound by HRP-conjugated secondary antibodies; the conjugated HRP catalyses a colorimetric reaction upon substrate addition that is read at 450 nm. The assay is described as lysate-free, high-throughput and sensitive.

The kit is intended for use with adherent cells and is reactive with human, mouse and rat samples. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance, and Crystal Violet whole-cell staining is provided as a second normalisation method to account for cell density and plating differences. The stated dynamic range is greater than 5000 cells. The kit is supplied in a 96-well microplate format and is stored at 4 °C with a stated shelf life of 6 months.

Rhodopsin is a G-protein coupled receptor that acts as the light-absorbing visual pigment of rod photoreceptor cells and mediates phototransduction in dim-light vision. Defects in rhodopsin are a cause of autosomal retinitis pigmentosa and congenital stationary night blindness, and the protein is therefore a relevant biomarker for studies of retinal disease within the signal transduction research area.
