The RFX3 Transcription Factor Activity Assay from Assay Genie is a non-radioactive DNA-binding ELISA kit for measuring the functional DNA-binding activity of RFX3 in nuclear and cell lysates. RFX3 belongs to the Regulatory Factor X family of transcription factors, recognizing X-box cis-regulatory motifs and governing cilia formation, cerebellar and forebrain patterning, and immune-related gene regulation.

Active RFX3 present in extracted nuclear samples binds a consensus double-stranded oligonucleotide immobilized on a microplate. A primary antibody directed against RFX3 is then added, followed by an HRP-conjugated secondary reagent and a substrate producing a measurable absorbance signal. The microplate kit provides 96 assays, with a recommended sample volume of approximately 20 uL per well for nuclear extracts. Reactivity has been validated for human, mouse, and rat samples, supporting translational studies in neurobiology, ciliopathy, and immunology.

Reagents should be stored frozen at -20 °C to preserve antibody and standard activity, with repeated freeze-thaw cycles of the standards minimized by aliquoting upon receipt. This is a research-use-only in vitro assay suitable for investigating RFX3-dependent transcriptional programs in developmental and disease models.
