RFWD2 (Phospho-Ser387)Colorimetric Cell-Based ELISA Kit

Assay Genie's colorimetric cell-based ELISA kit enables quantitative measurement of RFWD2 protein phosphorylated at serine 387 directly in cultured cells, without the need to prepare separate lysates. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures RFWD2 (Phospho-Ser387) and an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining can be used to correct for differences in cell density across wells.

The kit is reactive with human and mouse samples, with a dynamic range of > 5000 cells, and is supplied in a microplate format suitable for high-throughput screening of treatments, inhibitors (such as siRNA or chemical compounds), or activators on RFWD2 phosphorylation. RFWD2, also known as COP1, is an E3 ubiquitin-protein ligase implicated in cell cycle regulation, the DNA damage response, and protein degradation, and dysregulation of its phosphorylation has been linked to cancer progression, making the assay relevant to signal transduction research.
