Description

From Assay Genie, this Cell-Based ELISA Kit is designed to detect Retinoblastoma protein phosphorylation at Ser811 in cultured cells using a colorimetric method measured at 450 nm. The assay is reactive with human, mouse and rat samples and is suitable for adherent cells, with a dynamic range of > 5000 cells.

Working in an indirect ELISA format, the kit captures Retinoblastoma (Phospho-Ser811) with target-specific primary antibodies, while HRP-conjugated secondary antibodies catalyse a colour reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step is provided for determining cell density. The lysate-free, high-throughput workflow supports measurement of relative phosphorylated Retinoblastoma levels and screening of treatments, inhibitors (such as siRNA or chemicals) or activators on Retinoblastoma phosphorylation.

Associated target information: Gene Symbol RB1, Gene ID 5925, UniProt ID P06400, chromosomal location 13q14.2. The retinoblastoma protein is a tumour suppressor that regulates cell proliferation by controlling progression through the G1-phase restriction point, with cell cycle-dependent phosphorylation by Cdks modulating target binding and cell cycle progression.
