Description

Assay Genie offers this cell-based ELISA kit for the qualitative measurement of Retinoblastoma protein phosphorylated at Serine 795, providing a lysate-free, high-throughput workflow. The kit demonstrates reactivity with Human, Mouse, and Rat samples and uses a colorimetric detection method at 450 nm. Researchers can apply the assay to measure relative levels of phosphorylated Retinoblastoma in cultured cells and to screen the effects of treatments, inhibitors, or activators on Retinoblastoma phosphorylation.

The assay uses an indirect ELISA format in which phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bound to the Fc region of the primary antibody catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density so that absorbance readings can be normalized to cell amounts.

Designed for use with adherent cells, the kit has a dynamic range of > 5000 Cells. The target is the RB1 gene product, a tumor suppressor that regulates cell proliferation by controlling progression through the G1-phase restriction point, with phosphorylation at Serine 795 relevant to studies of cell signaling and cancer research.
