Description

Manufactured by Assay Genie, this Cell-Based ELISA is designed for the precise measurement of retinoblastoma protein levels in cell samples, offering high sensitivity and specificity across a range of research applications. The kit uses a colorimetric detection method measured at 450 nm and has a stated dynamic range of > 5000 Cells, with reactivity confirmed against human, mouse, and rat samples. It is suitable for use with adherent cell sample types and should be stored at 4°C with a stability of 6 months.

The assay is a convenient, lysate-free, high throughput format capable of detecting retinoblastoma protein expression profiles in cultured cells, allowing researchers to measure relative protein amounts and screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on retinoblastoma. Qualitative determination of retinoblastoma concentration is achieved through an indirect ELISA format, in which retinoblastoma-specific primary antibodies are captured and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

Because the Cell-Based ELISA is qualitative in nature, two normalization methods are provided. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method can be used to determine cell density. Results are analysed by normalizing the absorbance values to cell amounts, which adjusts for plating differences.
