Ret Colorimetric Cell-Based ELISA Kit

From Assay Genie, this in-cell ELISA format provides a lysate-free, high-throughput approach for the qualitative measurement of Ret protein expression in cultured cells. The assay uses an indirect ELISA configuration, in which Ret-specific primary antibodies are paired with HRP-conjugated secondary antibodies to generate a colorimetric signal at 450 nm upon substrate addition.

Designed for use with adherent cell samples, the kit is reactive with Human, Mouse and Rat samples and supports a dynamic range of > 5000 cells. An included monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance values, while a Crystal Violet whole-cell staining step allows cell density to be taken into account after the HRP readout, helping to adjust for plating differences. The kit is supplied in a 96-well microplate format and is intended to be stored at 4 °C, with a stability of 6 months under those conditions.

RET is a proto-oncogenic receptor tyrosine kinase, and dysregulation of RET signaling has been linked to multiple cancers, making this assay relevant to studies of cancer biology and drug discovery. The kit can be applied both for measuring relative Ret levels in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on Ret expression.
