Description

From Assay Genie, this colorimetric Cell-Based ELISA assay is designed for the detection of phosphorylated Rel at serine 503 in cell lysates and cell culture supernatants, and is reactive with human samples in adherent cell culture. The kit provides a qualitative, high-throughput, sensitive and lysate-free workflow with a dynamic range of > 5000 cells for measuring Rel protein phosphorylation and expression profiles in cultured cells, and can be used to screen the effects that various treatments, inhibitors (such as siRNA or chemicals) or activators have on Rel phosphorylation.

The assay is based on an indirect ELISA format in which target-specific primary antibodies capture phosphorylated Rel, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition, read at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance, and following the HRP measurement, Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance values to be normalised to cell amounts.

Classed as a phospho-specific Cell-Based ELISA within the epigenetics and nuclear signalling research area, the kit supports studies of cellular signalling pathways in which phosphorylation of serine 503 is implicated, as well as investigations relevant to diseases such as cancer, inflammatory disorders and neurodegenerative conditions.
