Rat Vitamin K-dependent protein C (Proc) ELISA Kit

Designed for the quantitative measurement of rat Vitamin K-dependent protein C (Proc), this sandwich, double-antibody ELISA from Assay Genie accepts serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. An antibody specific to the target protein is pre-coated onto the wells of a microplate, followed by a biotin-conjugated detection antibody and avidin conjugated to Horseradish Peroxidase (HRP). TMB serves as the substrate, with a sulfuric acid stop solution, and the optical density is read at 450 nm ± 2 nm.

Performance characteristics include a detection range of 0.312-20 ng/mL and a sensitivity of 0.156 ng/mL, with an intra-assay CV of 4.9% and inter-assay CV of 7.5%. Linearity has been reported in serum (92-102% at 1:2; 81-91% at 1:4; 86-95% at 1:8; 106-115% at 1:16), EDTA plasma (101-111% at 1:2; 94-103% at 1:4; 111-120% at 1:8; 112-121% at 1:16) and heparin plasma (96-106% at 1:2; 96-107% at 1:4; 100-110% at 1:8; 97-106% at 1:16). Spike-and-recovery performance shows averages of 96% (90-102% range) in serum and 98% (92-104% range) in plasma.

Protein C (UniProt P31394, NCBI Gene ID 25268) is a vitamin K-dependent serine protease (EC 3.4.21.69) with a molecular weight of 55.2 kDa. It regulates blood coagulation by inactivating factors Va and VIIIa in the presence of calcium ions and phospholipids, and exerts a protective effect on endothelial cell barrier function. The assay recognises natural and recombinant rat Vitamin K-dependent protein C and is for research use only.
