Description

Assay Genie provides a sandwich-format ELISA kit for the quantitative measurement of vascular endothelial growth factor B (VEGF-B) in rat-derived samples, using a double-antibody configuration with colorimetric readout. The kit is suited to serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid sample types. VEGF-B is a growth factor involved in angiogenesis and the regulation of blood vessel formation and is relevant to research into cancer, cardiovascular disorders, and ischemic diseases.

Performance specifications include a sensitivity of 9.375 pg/mL and a standard curve range of 15.625–1000 pg/mL. Dilutional linearity has been verified in serum (90–103% at 1:2, 83–101% at 1:4, 82–97% at 1:8), EDTA plasma (86–104%, 84–100%, 83–95%), and heparin plasma (85–103%, 83–96%, 85–99%). Spike-and-recovery ranges are 85–103% in serum (average 95%), 85–101% in EDTA plasma (average 93%), and 87–100% in heparin plasma (average 93%). Reagents are stored at 2–8°C for 12 months.

The workflow equilibrates reagents and TMB substrate to room temperature on the pre-coated plate, followed by a 90-minute primary incubation at 37°C. A biotin-labeled detection antibody is then applied for 60 minutes, HRP-streptavidin for 30 minutes, and TMB substrate in the dark for 10–20 minutes, before stopping the reaction and reading absorbance at 450 nm. Sample preparation notes cover serum (clot, 1000 × g, 20 minutes), plasma (anticoagulant tubes, 1000 × g, 15 minutes at 2–8°C), tissue homogenate in PBS with protease inhibitors, cell culture supernatant clarified by centrifugation, and cell lysates prepared with lysis buffer. The kit is supplied with a pre-coated microplate, lyophilized standard, biotin-labeled antibody, HRP-streptavidin conjugate, TMB substrate, sample, antibody and SABC dilution buffers, stop solution, wash buffer, plate sealer, and a technical manual.
