Description

The Assay Genie Rat VAP-1 ELISA Kit is a sandwich (double antibody) enzyme-linked immunosorbent assay intended for the quantitative measurement of Vascular Adhesion Protein 1 (VAP-1), also referred to as membrane primary amine oxidase (Aoc3), in rat-derived samples. The kit has a stated sensitivity of 0.469 ng/mL and a working range of 0.781–50 ng/mL, and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. According to the manufacturer, VAP-1 is implicated in leukocyte adhesion and migration to sites of inflammation and is associated with conditions such as arthritis, atherosclerosis, and diabetic complications.

The assay is run on a pre-coated 96-well microplate. Standards and samples are incubated at 37°C for 90 minutes, followed by a biotinylated detection antibody (37°C, 60 minutes), HRP-Streptavidin SABC (37°C, 30 minutes), and TMB substrate developed in the dark for 10–20 minutes. The reaction is stopped and absorbance is read at 450 nm. Reported dilution linearity (n = 5) for serum is 88–105% at 1:2, 83–99% at 1:4, and 80–95% at 1:8; for EDTA plasma, 86–101%, 83–101% and 83–99%; and for heparin plasma, 89–104%, 84–98% and 82–98%. Mean spike recoveries are 93% for serum, 91% for EDTA plasma, and 92% for heparin plasma.

Kit components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a Bradford Reagent for total protein normalisation. Components are shipped on ice packs and stored at 2–8°C; the kit is supplied for research use only.
