Description

From Assay Genie, this sandwich ELISA kit employs a double-antibody format with colorimetric detection to quantify uromucoid levels in rat samples. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit recognizes uromucoid, also referred to as uromodulin, Tamm-Horsfall glycoprotein, THP, or UMOD. Sensitivity is 1.875 ng/mL with a working range of 3.125-200 ng/mL, and components are stored at 2-8°C for 12 months.

The kit includes pre-coated microplate strips, lyophilized standard, biotin-labeled detection antibody (100X), HRP-streptavidin conjugate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer (25X), plate sealer, and a technical manual. The assay workflow uses sequential incubations at 37°C, with biotin-labeled detection antibody binding, HRP-streptavidin binding, and TMB substrate development in the dark, followed by absorbance measurement at 450 nm after stop solution addition.

Reported linearity across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions (n=5) was 88-103%, 86-105%, and 85-103% for serum; 87-96%, 82-100%, and 85-98% for EDTA plasma; and 82-99%, 81-94%, and 81-98% for heparin plasma. Recovery ranges were 87-100% (average 94%) for serum, 86-99% (average 93%) for EDTA plasma, and 85-98% (average 90%) for heparin plasma. Uromucoid is associated with kidney function, and elevated levels have been linked to kidney disease, urinary tract infections, and kidney damage, supporting its use as a biomarker for renal research.
