Rat uPA/Urokinase-Type Plasminogen Activator ELISA Kit

From Assay Genie, this sandwich ELISA with double-antibody detection uses a colorimetric readout to measure uPA (Urokinase-Type Plasminogen Activator) levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. uPA is a key enzyme in fibrinolysis, cell migration, and tissue remodeling, and its dysregulation has been implicated in cancer, inflammation, and cardiovascular diseases.

The assay has a reported sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL. Linearity in serum (n = 5) was 88–99% at 1:2, 88–100% at 1:4, and 80–100% at 1:8; EDTA plasma (n = 5) gave 85–104%, 84–101%, and 86–91%; heparin plasma (n = 5) gave 86–103%, 83–97%, and 82–95%. Average recovery was 94% for serum and EDTA plasma and 96% for heparin plasma, with recovery ranges of 88–105%, 87–100%, and 87–104% respectively.

Unopened kit components are stored at 2–8°C for 12 months. The protocol includes primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, and TMB colour development in the dark for 10–20 minutes, with absorbance read at 450 nm after adding stop solution.
