Rat TSH ELISA Kit

A competitive ELISA from Assay Genie, coated with antigen and based on a double-antigen sandwich workflow, is designed for the quantitative measurement of Thyroid Stimulating Hormone (TSH) in rat samples. The kit uses a microplate pre-coated with a specific antigen, where standards and appropriately diluted samples are added, followed by a biotin-labelled antigen to form a sandwich complex. HRP-conjugated Streptavidin and TMB substrate produce a colorimetric reaction, with absorbance measured at 450 nm; signal intensity is directly proportional to analyte concentration. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay has a stated sensitivity of 0.75 ng/mL and a working range of 1.25-80 ng/mL. Linearity has been characterised for serum (91-105%, 88-103% and 88-105% at 1:2, 1:4 and 1:8 dilutions respectively), EDTA plasma (82-101%, 89-101% and 83-99%) and heparin plasma (88-100%, 82-94% and 85-100%). Recovery ranges from 89-99% in serum (average 95%), 85-101% in EDTA plasma (average 91%) and 88-105% in heparin plasma (average 99%).

The protocol calls for a 45-minute sample and biotin-labelled antibody incubation at 37 °C, five washes with a 1-minute soak, TMB colour development in the dark for 10-20 minutes, stop solution (colour changes from blue to yellow), and absorbance measurement at 450 nm. The kit should be stored sealed at 2-8 °C and not cryopreserved.
