Description

From Assay Genie, this Sandwich ELISA Kit is designed for the precise quantification of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) levels in rat samples. The kit has a stated sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL, making it suitable for measuring TRAIL in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody sandwich format with TMB colour development and is read at 450 nm. Kits should be stored at 2-8°C for up to 12 months.

TRAIL is described as a key protein in apoptosis, or programmed cell death, and has been implicated in disease states including cancer, autoimmune disorders, and inflammatory conditions, making accurate measurement valuable for research into cell death pathways and potential therapeutic strategies. Reported linearity for serum (n=5) is 86-104% at 1:2, 85-95% at 1:4, and 80-95% at 1:8 dilution, with EDTA plasma at 92-105%, 85-96%, 85-100% and heparin plasma at 91-102%, 88-99%, 80-95%. Reported recovery averages are 96% for serum (range 87-101%), 95% for EDTA plasma (88-105%), and 96% for heparin plasma (89-103%).

The assay procedure includes reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction read at 450 nm. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate, with additional support available from the Assay Genie technical team for other sample types.
