Description

A sandwich ELISA from Assay Genie is designed for the quantitative detection of TNNT3 (Troponin T, fast skeletal muscle) levels in rat samples. The kit uses a double-antibody sandwich format with colorimetric detection and is suitable for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. TNNT3 is a critical component of the troponin complex in fast skeletal muscle, where it plays a key role in muscle contraction and regulation; abnormal levels have been implicated in muscle disorders, making it a valuable biomarker for skeletal muscle research.

The assay has a sensitivity of 18.75 pg/mL and a standard range of 31.25-2000 pg/mL. Reported linearity (n = 5) at 1:2, 1:4 and 1:8 dilutions is 86-101%, 84-97% and 82-99% for serum; 86-104%, 83-93% and 84-94% for EDTA plasma; and 89-94%, 86-95% and 81-96% for heparin plasma. Average recovery is 97% in serum, 95% in EDTA plasma and 98% in heparin plasma. The kit is stored at 2-8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation for 60 minutes, HRP-Streptavidin (SABC) incubation for 30 minutes, TMB substrate colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution. Sample preparation guidance includes centrifuging serum at 1000 × g for 20 minutes, plasma at 1000 × g for 15 minutes at 2-8°C, and cell culture supernatant at 2500 rpm for 5 minutes.
