TNF-beta, more formally known as lymphotoxin-alpha (LTα), is a soluble cytokine released mainly by activated Th1 lymphocytes and lymphoid-tissue inducer cells. It signals through TNFR1 and the LTβ receptor to drive lymphoid organogenesis, splenic microarchitecture, and germinal-centre maintenance, so quantifying circulating LTα in rat models is particularly relevant to studies of mucosal immunity, dextran-sulphate-induced colitis, cardiac allograft rejection, and chronic inflammation, where LTα levels tend to track lymphocytic infiltration rather than macrophage-derived TNF-alpha activity.

The Assay Genie kit is configured for a complete microplate and tolerates 50 uL of serum or plasma per well, while cell-culture supernatant, cell lysate, and tissue lysate are loaded at 100 uL. The matched capture and detection antibodies show no significant cross-reactivity with rat TNF-alpha, IL-1 beta, IL-6, or related analogues, giving clean discrimination between the two TNF-family ligands during multiplex-style cytokine profiling. Reconstituted standards remain usable for roughly 30 days under refrigerated storage and for up to 12 months at -20 °C when aliquoted to limit freeze-thaw damage, and the supplied TMB substrate and stop solution are light-sensitive and should be returned to the fridge immediately after each run to preserve the chromogenic signal across plates.
