Description

This sandwich ELISA kit from Assay Genie is configured for the quantitative detection of rat TNF-alpha using a double-antibody, colorimetric workflow and is validated for serum and plasma samples. The assay has a reported sensitivity of 2.344pg/ml across a measuring range of 3.906-250pg/ml, with intra-assay CV <8% and inter-assay CV <10%. Kits should be stored at 4°C for 6 months and are intended for research use only.

Spike-and-recovery in serum (n=5) was 87-105% (average 95%), in EDTA plasma 90-100% (average 95%) and in UFH plasma 89-100% (average 94%). Dilution linearity across 1:2, 1:4 and 1:8 was 85-103%, 85-101% and 85-97% for serum; 83-99%, 82-98% and 84-96% for EDTA plasma; and 81-99%, 81-100% and 83-97% for UFH plasma. The target analyte is rat TNF-alpha (UniProt P16599, gene symbol Tnf), a 25,806 Da homotrimeric cytokine of the tumour necrosis factor family. It is mainly secreted by macrophages, binds the receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR, and is implicated in pyrogenic responses, induction of cachexia, inflammatory signalling and apoptosis. The rat gene is also known by synonyms including Tnfa, RATTNF and TNF-alpha, with NCBI Gene ID 24835.
