About TIMP1 and Assay Use

Quantitative measurement of tissue inhibitor of metalloproteinases 1 (TIMP1) in rat specimens is the purpose of this microplate-based ELISA kit supplied by Assay Genie. TIMP1 is an approximately 28 kDa secreted glycoprotein that regulates extracellular matrix remodeling by forming 1:1 stoichiometric complexes with active matrix metalloproteinases, including MMP-1, MMP-2, MMP-3, MMP-7, and MMP-9. In rat models, altered TIMP1 expression is investigated in settings such as carbon tetrachloride- or bile duct ligation-induced hepatic fibrosis, post-infarction myocardial remodeling, diabetic nephropathy, bleomycin-induced pulmonary fibrosis, and chemical carcinogenesis protocols.

Kit Format and Specimen Compatibility

The kit provides reagents for 96 assays configured on a strip-well microplate and is intended for use with serum- or plasma-type specimens as well as tissue homogenates, cell lysates, and cell culture supernatants. Reagents should be equilibrated to room temperature before opening; reconstituted standards and detection antibody are refrigerated and used within the stability window stated in the kit insert. Unused strips are returned to the foil pouch with desiccant and refrigerated between runs to preserve activity.

Workflow Notes

Standards and samples are typically diluted into the linear portion of the standard curve, and assay buffer, wash concentrate, and substrate solution are supplied either ready-to-use or as concentrated stocks as listed on the enclosed protocol sheet. For studies pairing TIMP1 quantitation with downstream zymography, immunohistochemistry, or western blot analysis of MMP expression in the same rat cohort, the kit provides a quantitative readout that complements those orthogonal readouts.
