Description

This Sandwich ELISA from Assay Genie is designed for the quantitative measurement of Thrombin-antithrombin complex (TAT) in rat samples. It uses a Double Antibody format with colorimetric detection and is reactive with rat serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported performance figures are a sensitivity of 9.375 pg/mL and a standard range of 15.625-1000 pg/mL. The kit is stored at 2-8°C for 12 months. Linearity at dilutions of 1:2, 1:4 and 1:8 is reported as: Serum (n=5) 90-102%, 88-103%, 87-101%; EDTA Plasma (n=5) 82-100%, 86-101%, 85-96%; Heparin Plasma (n=5) 81-87%, 86-92%, 92-98%. Recovery is reported as: Serum (n=5) 85-103%, average 92%; EDTA Plasma (n=5) 87-105%, average 96%; Heparin Plasma (n=5) 85-103%, average 94%. The assay procedure involves equilibrating reagents to room temperature, primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution.

TAT is described as an important biomarker reflecting the balance between thrombin generation and inhibition in the blood coagulation process. Monitoring TAT levels is described as useful in studying thrombotic disorders, including deep vein thrombosis and pulmonary embolism, and in assessing the effectiveness of anticoagulant therapy, making this kit an essential tool for researchers investigating coagulation pathways and developing treatments for thrombotic diseases in rat models.
